Course Objectives

The course provides participants with updated background information and a practical approach to the study of clinically relevant mechanisms of antimicrobial resistance. This edition of the workshop will focus on Pseudomonas aeruginosa expressing extended spectrum beta-lactamases or carbapenemases with altered outer membrane permeability, mutations in QRDR of the DNA gyrase, mutations in the mismatch repair system genes and hyperexpressing efflux pumps or the chromosomal beta-lactamase AmpC.

Laboratory sessions

  • Antibiotic susceptibility tests: disk diffusion and microdilution.
  • Typing of clinical isolates: pulsed field gel electrophoresis, REP-PCR and genotyping array.
  • Hypermutation and antimicrobial resistance: plasmidic DNA preparation and characterisation, preparation of competent bacterial cells, artificial transformation assays and mutation rate.
  • Analysis of chromosomal resistance genes expression: RNA preparation and RT-PCR.
  • Contribution of acquired beta-lactamases to antimicrobial resistance: chromosomal DNA preparation, amplification of resistance genes by PCR, conjugation and spectrophotometric assay for beta-lactamase activity.
  • Identification of outer membrane proteins: SDS-PAGE, Western blot and tandem mass spectrometry.
